I have two client tunnels set up in i2ptunnelmgr, listening on the same port but on two completely different interfaces (and therefore ip addresses). In my case one is listening on 127.0.0.1:4444 and another on 10.x.x.x:4444, but the actual ips and ports don't really matter. Just create any two client tunnels and have them listen on the same port (but with different ips) and you'll get the "Duplicate port" warning.
Afaik there should be no conflict between the two. I guess it's just an error in the configuration validation system, and there seems to be no interference between the two tunnels (each one appears to work as expected).

Thanks for the report. Yes, it's only a warning, and it only compares port number.

I can fix it to compare interface as well.
